A: Yes you should be able to. I have taken a 1DXii, 6D, 100-400, 24-70, and 70-200 f2.8 w sundry accessories

A: Not easily, you could put a bladder in the back of the bag but because it is meant for camera gear, you would have to take extra measures to ensure your bladder doesn't leak.

Q: 3. What can fit in the front of the bags main zipper compartment? It states in the description that it has two zippered panels which I see, but how deep is that pocket and what can it hold?

A: The outside front pocket has one zipper. As Mindshift says. Front zippered pocket fits 6 liters of extra gear to extend your adventure. The inside lid has two zippers. A: Yes this bag could fit the Canon 1Dx Mark II and a Canon 400mm f/5.6L lens. They may not be able to fit attached, but it has the capacity to fit both.

Q: 5. Can this hold a 16" macbook pro and a pro body (or body + battery grip) standing? Will it work with/out a lens attached?

A: Yes, when closed with a laptop and a non-pro body, the laptop (and items in other the pouches in the laptop area) do not press against a non-pro body. A: The waist belt on the Backlight 18L cannot be removed, but it can be stowed by pulling the belt tabs back and clipping it to the lower front portion.
