If you use shutter button (half press), and then, keeping it half pressed, tweak manually it may work.If you use AF-C the camera will override any tweak/s.If you set Direct Manual Focus (DMF), it will start as AF-S and then go to manual focus mode where you can tweak the focus -- it will stay in manual focus as long as you are in half press shutter button. DMF is even more helpful if you have set Peaking Level and Color.
