It's a nice feature for those who prefer manual focus or wish to critically focus their scene, but if you wish to keep it in autofocus mode at all times, that's okay. I rarely use this feature myself.

A: As long as you have three separate Tilta Wireless Thumb Controller for DJI RS 2/RS 3 PRO modules attached and connected to the appropriate thumb controller, it should work. Please keep in mind to change each channel each to a different channel, preferably not the next channel over, as they could be a similar signal and there might be some confusion between the connections.
